I just re-started a game yesterday on a fresh install of Windows 7 and have selected Captain America, Ms. Marvel, Spider-Woman and Iron Man.  I completed the game once on Vista but I noticed that everyone I unlocked was available at the beginning of the game.  I had reloaded an old save from after I finished the game the first time and I think that unlocked the extra characters.  I still haven't unlocked Silver Surfer yet.

I've been primarily controlling Spider-Woman and she does pretty well with her ranged attack (venom blast?).
